Abstract Objective To establish the method for determination of Vancomycin in human plasma by high performance liquid chromatography. Methods High performance liquid chromatography with norvancomycin as internal standard was used for detection. The liquid chromatography was performed on Kinetex C18(100 mm×4.6 mm, 2.6 μm), and acetonitrile-0.05 mol/L potassium dihydrogen phosphate buffer (pH=3.2) was used as the mobile phase to gradient elute, the flow rate was 1.0 ml/min, the column temperature was 40℃, and the detection wavelength was 220 nm. The plasma sample was precipitated with Zinc Sulfate and injected with a volume of 10 μl. Results The linear range of Vancomycin was 2.22-111.11 μg/ml (R2=0.9995, n=6), intra-day relative standard deviation<4.86%, intra-day relative standard deviation<3.72%, the absolute recovery was over 94.6%. Conclusion High performance liquid chromatography is sensitive and accurate, which can be used in clinical detection of plasma Vancomycin concentration, guide clinical individualized drug administration, improve the efficacy and reduce the incidence of adverse reactions.
