Abstract Objective To investigate the expression and significance of minichromosome maintenance protein 2 (MCM2) in head and neck squamous cell carcinoma (HNSCC). Methods The Gene Expression Omnibus (GEO) databases were adopted to compare the expression of MCM2 in HNSCC with normal tissues, and the UALCAN database was used to analyze the expression of MCM2 in HNSCC and normal tissues in different conditions. The association of MCM2 with immune cells was analyzed by the TIMER database. Linked Omics and STRING databases were used to study genes closely related to MCM2. Finally, GSCALite database was used to analyze drug targets of the related key genes. Results The expression of MCM2 in HNSCC was higher than that in normal tissues, the difference was statistically significant (P<0.05). The expressions of MCM2 in HNSCC of different age groups, gender, tumor grades, clinical stages and lymphatic metastasis were higher than that in normal tissues, the expression of MCM2 in human papilloma virus (HPV) positive patients was higher than that in HPV negative patients, the difference was statistically significant (P<0.05). The expression of MCM2 in HNSCC was positively correlated with the infiltration level of immune cells (P<0.05). GSCALite database analysis showed MCM2 was regulated by nPK76 -II -72 -1, Navitoclax, GSK1070916 and I -BET -762. Conclusion MCM2 was highly expressed in HNSCC, which could be used as a new target for diagnosis and drug treatment of HNSCC.
