Abstract Objective To investigate the effect of 70% ethanol extract of Fructus Gardeniae Leaves on the melanin formation and tyrosinase activity of B16 melanoma cells.Methods The B16 melanoma cells were divided into blank group,model group,positive control group and 70% ethanol extract of Fructus Gardeniae Leaves group (low concentration group [100 mg/L],medium concentration group [200 mg/L] and high concentration group [400 mg/L]).The B16 melanoma cells were addressed with 70% ethanol extract of Fructus Gardeniae Leaves and induced by α-melanocytestimulating hormone (α-MSH) for 72 h.The proliferation activity of the cells was detected by MTS,and the content of melanin and the activity of tyrosinase in the B16 melanoma cells were determined by enzyme linked immunosorbent assay..Results The 70% ethanol extract of Fructus Gardeniae Leaves with concentration of 100-400 mg/L had no effect on the proliferation of B16 melanoma cells.The content of melanin and tyrosinase activity in B16 melanoma cells in model group were significantly higher than those in blank group,with highly statistically significant differences (P<0.001).The content of melanin and tyrosinase activity in B16 melanoma cells in the 70% ethanol extract of Fructus Gardeniae Leaves were lower than those in the model group,and the differences were statistically significant (P<0.05).The content of melanin and tyrosinase activity in B16 melanoma cells in the positive control group were significantly lower than those in the model group,and the differences were highly statistically significant (P<0.001).Conclusion The 70% ethanol extract of Fructus Gardeniae Leaves can effectively inhibit the generation of α-MSH-induced melanin in B16 melanoma cells,and the mechanism may be related to the inhibition of tyrosinase activity
