基于网络药理学的葛根降糖机制研究

Abstract Objective To study the main active components and targets of Radix Puerariae in hypoglycemic action based on network pharmacology,analyze and find the pathway of Radix Puerariae to exert its pharmacodynamic effect and to understand its synergistic mechanism of multi-target and multi-pathway.Methods The related targets of Radix Puerariae were obtained by using the pharmacology database and analysis platform of traditional Chinese medicine system(TCMSP),and the related pathways and functional annotation information were obtained and analyzed by protein-protein interaction database platform and gene function analysis platform.By protein protein interaction networks and network analysis software "compounds-target-signal path-function" network to further determine the target features,through the molecular complex testing plug-ins,combined with its function and pathways for the clustering of proteinprotein interaction networks,different modules and each module of key genes.The binding of small molecular components of pueraria root and their targets was verified by molecular docking.Results Four components including formononetin,sitosterol,3′-methoxydaidzein,daidzein-4′,7-diglucoside were screened.Three main targets CYP17A1,NR3C1 and HTR2C were obtained.Conclusion Through the comprehensive analysis of target function,pathway and protein-protein interaction network,three target modules were obtained.By analyzing their possible mechanism of action in hypoglycemia and their research status,it can provide a certain basis for the efficient utilization of Radix Puerariae.
