Objective To establish an ultra-high performance liquid chromatography-andem mass spectrometry (UPLCMS/MS) method for the content determination of rutin in Xiakucao Tablets.Methods UPLC-MS/MS method was used and the chromatographic column was ACQUITY UPLC RBEH C18 column (2.1 mm×100 mm, 1.7 μm), the mobile phase was methanol-aqueous solution(47：52).The mass-charge ratio(m/z)of 300.1 ion was determined by using an electrospray ionization (ESI) in positive ionization mode.Results The established method was able to rapidly detect the rutin content in Xiakucao Tablets with a good linear relationship (r=0.9993) within the concentration range of 10.78-172.48 ng/ml,and the average recovery rate was 99.44%.The method had good repeatability and precision.Conclusion The method has strong specificity, rapid sensitivity and is suitable for the content determination of rutin in Xiabancao Tablets.
