Application of PCR fluorescence probe method combined with DNA micro-display chip method in the diagnosis and treatment of pulmonary tuberculosis |

Abstract Objective To explore the application effect of PCR fluorescence probe method combined with DNA microdisplay chip method in the diagnosis and treatment of pulmonary tuberculosis.Methods A total of 128 patients with pulmonary tuberculosis admitted to Shangrao Second People′s Hospital from January to October 2020 were selected as the observation group.Meanwhile,40 patients without pulmonary tuberculosis were selected as the control group.Morning sputum of the two groups were collected for detection of Mycobacterium tuberculosis using acid-fast staining of sputum smear,serum tuberculosis antibody and PCR fluorescent probe.Then DNA micro-display chip method was used to detect drug resistance of Mycobacterium tuberculosis.The diagnostic efficiencies of the three methods were compared,and the results of rifapicin isoniazid resistance detection by DNA microarray chip method were analyzed.Results The sensitivity,specificity,positive predictive value and accuracy of PCR fluorescence probe in the diagnosis of tuberculosis were higher than those of acid-fast staining of sputum smear and serum tuberculosis antibody testing,with statistically significant differences(P<0.05).DNA micro-display chip method found 45 cases sensitive to rifampicin,11 cases resistant to rifampicin,44 cases sensitive to isoniazid,12 cases resistant to isoniazid,and 5 cases resistant to both drugs.Conclusion PCR fluorescence probe method combined with DNA micro-display chip method can not only increase the detection rate of Mycobacterium tuberculosis,but also quickly detect the drug resistance of Mycobacterium tuberculosis,and the diagnosis and drug resistance results can be obtained within 48 hours,thereby assisting in early diagnosis and development of effective treatment plans,which is of great significance for improving the prognosis of patients with pulmonary tuberculosis.
