Abstract: Objective To investigate the effect of capillary hemoglobin electrophoresis combined with mean corpuscular volume (MCV) in couples of childbearing age β-thalassemia screening.Methods From August 2019 to September 2020,1508 couples of childbearing age (3016 people) who did pre-pregnancy β-thalassemia screening in Maternal and Child Health Hospital of Qingxin District,Qingyuan City were selected as the research objects.Fasting venous blood was collected,and the distribution of hemoglobin (Hb) was detected by capillary hemoglobin electrophoresis and MCV was detected by blood cell analyzer for β-thalassemia screening.Polymerase chain reaction/reverse dot blot (PCR/RDB) was used for gene diagnosis.The gene diagnosis result was took as the gold standard,the diagnostic efficacy of capillary hemoglobin electrophoresis and MCV alone and combined in screening β-thalassemia in couples of childbearing age was analyzed,and the Hb distribution of different clinical types of β-thalassemia was analyzed.Results The results of gene diagnosis showed that 174 of 3016 samples were β-thalassemia gene positive,with a positive rate of 5.77%,including 168 cases of mild type and 6 cases of intermediate type.The content of Hb-A in patients with intermediate βthalassemia was lower than that in patients with mild β-thalassemia,the difference was statistically significant (P<0.05).The sensitivity,specificity and accuracy of β-thalassemia screening by capillary hemoglobin electrophoresis combined MCV were 95.98%,91.86% and 92.09% respectively.Conclusion The combination of capillary hemoglobin electrophoresis and MCV detection for prenatal β-thalassemia screening of couples of childbearing age can improve the sensitivity and accuracy of screening to a certain extent,and has certain guiding significance for their clinical typing.
