Establishment of non-alcoholic fatty liver model in type 2 diabetic rats

Abstract:Objective To establish a non-alcoholic fatty liver model in rats with type 2 diabetes, and to explore the effect of type 2 diabetes on the liver of rats. Methods Forty healthy male SD rats were randomly divided into normal control group (NC) and diabetic group (DM), with 20 rats in each group. Type 2 diabetic rats in DM group were induced by intraperitoneal injection of streptozotocin (STZ, 35 mg/kg) combined with high-glucose and high-fat diet. After modeling, blood glucose, triacylglyceride (TG), total cholesterol (TC), alanine aminotransferase (ALT), aspartate aminotransferase (AST), liver index, insulin sensitive index (ISI), Homeostasismodel assessment insulin resistance index (HOMA-IR) related indicators were detected. Liver tissue of rats was stained with HE and its pathological changes were observed. Results At 6, 7, 8, 9, 10 weeks after modeling, the body weight of DM group was lower than that of NC group, and blood glucose was higher than that of NC group, the differences were statistically significant (P<0.05). TG, TC, ALT and liver index in DM group were higher than those in NC group, the differences were statistically significant (P<0.05). There was no significant difference in AST between the two groups (P>0.05). 6, 10 weeks after modeling, the fasting insulin (FINS), HOMA-IR of DM group were higher than those of NC group, ISI was lower than that of NC group, the differences were statistically significant (P<0.05). Under the light microscope, the normal liver cells in NC group were orderly, the liver lobules were normal, the cytoplasm was round and uniform, and there was no infiltration of steatosis and inflammatory cells. In DM group, liver cells were improperly organized, fatty degeneration, liver cell edema, infiltration of inflammatory cells, partial spots and local necrosis. The activity score of non-alcoholic fatty liver disease (NAS) in DM group was higher than that in NC group, and the difference was statistically significant (P<0.05). Conclusion Intraperitoneal injection of low-dose STZ combined with high-sugar and high-fat feeding can induce a non-alcoholic fatty liver model of type 2 diabetes in rats.
