Effect of tetrahydroxystilbene glucoside on the expression of Nalp3,Caspse-1, interleukin-18 in rats with cerebral ischemia-reperfusion injury

Abstract:Objective To explore the effect of tetrahydroxystilbene glucoside(TSG)on the expression of Nalp3,Caspse-1,interleukin-18 (IL-18) in rats with cerebral ischemia-reperfusion injury. Methods Sixty SD rats were randomly divided into five groups, namely sham operation group, model group, TSG low-dose group, TSG medium-dose group, TSG highdose group, with 12 rats in each group. The bilateral common carotid arteries were clamped for 2 h, and the arterial clamps were removed to restore blood perfusion to prepare the animal model of cerebral ischemia-reperfusion injury.The sham operation group was anesthetized and bilateral common carotid arteries were separated without clamping. TSG low, medium and high dose groups were given TSG 30, 60, 90 mg/kg, intraperitoneal injection once a day for 7 consecutive days. The sham operation group and model group were given the same dose of normal saline. On the 8th day after the operation, the rats were anesthetized, and the related specimens were reserved. The histomorphological changes of HE staining hippocampus were observed by light microscope. The brain water content of rats in each group was calculated by dry and wet weight method. The expressions of Nalp3 mRNA, Caspase-1 mRNA and IL-18 mRNA in hippocampus were detected by RT-PCR. Results HE staining showed that the hippocampal nerve cells in the model group were seriously damaged. The damage of hippocampal nerve cells in TSG medium and high dose group were significantly reduced. The brain water content of rats in the model group and the expressions of Nalp3 mRNA,Caspase-1 mRNA and IL-18 mRNA in the hippocampus were higher than those in the sham operation group, and the differences were statistically significant (P<0.05). The brain water content and the expressions of Nalp3 mRNA,Caspase-1 mRNA and IL-18 mRNA in the hippocampus of rats in the middle and high-dose TSG groups were lower than those in the model group, and the differences were statistically significant (P<0.05). Conclusion TSG can inhibit the expression of Nalp3, Caspse-1, and IL-18 in the hippocampus of rats with cerebral ischemia-reperfusion injury, reduce the inflammation cascade, and have a protective effect on nerve cells.
