Abstract:Objective To investigate the application effect of injection platelet-rich fibrin combined with Bone marrow mesenchymal stem cells (BMSCs) on sciatic nerve injury.Methods A total of 6 cases 10-day-old SD suckling rats of either sex were cultured and purify BMSCs group for later use,and 24 2-month-old SD rats half male and half female of either sex were selected,and 24 cases 2-month-old SD rats were randomly divided into observation group and control group,each had 12 cases.A rat sciatic nerve injury model was established by modified extrusion injury method.BMSCs suspension 250 μl and autologous platelet-rich fibrin 250 μl were locally injected into the observation group and the injury site,and 500 μl of normal saline was locally injected into the control group.Then the changes of sciatic nerve function index before and after intervention were compared between the two groups.The result of transmission electron microscope images of sciatic nerve axons of rats were analyzed 14 days after treatment of the two groups.The changes of wet quality of gastrocnemius muscle in 14 days after intervention in the two groups were counted.Results Before treatment,there was no significant difference in sciatic nerve function index between the two groups (P>0.05);after treatment,the sciatic nerve function index of the observation group was lower than that before treatment and of the control group,the difference was statistically significant (P<0.05); after 14 days of treatment,the rat sciatic nerve transmission electron microscope showed that the ratio of myelin axons in the observation group was higher than that in the control group,the wet mass of the gastrocnemius in the observation group was higher than that in the control group,and the recovery rate of the wet mass of the gastrocnemius in the observation group was higher than that in the control group,the differences were statistically significant (P<0.05).Conclusion For sciatic nerve injury rats,the use of autologous injection platelet-rich fibrin combined with BMSCs for treatment can effectively improve sciatic nerve function,so it can promote nerve repair and improve smooth muscle and vascular regeneration.
