Expression and role of TDP-43 and LC3 in polymyositis and sporadic inclusion body myositis

Abstract:Objective To explore the expression and role of TDP-43 and LC3 in polymyositis (PM) and sporadic inclusion body myositis (IBM). Methods From March 2018 to September 2019, the moderately affected muscles of PM(26 cases)and IBM (26 cases) who were admitted to our hospital were selected for biopsy, and the electromyographic puncture points were avoided. The muscle tissue of the PM patients was taken as the PM group, and the muscle tissue of the IBM patients was taken as the IBM group. The differences of TDP-43, LC3 positive expression rate and protein content between PM group and IBM group by immunohistochemistry, Western blot and other methods were analyzed,and then the expression and role of TDP-43, LC3 in PM and IBM was explored. Results The results of immunohistochemistry showed that the positive expression of TDP-43 and LC3 in the muscle tissue of the IBM group was higher than that of the PM group; the results of Western blot showed that the protein contents of TDP-43 and LC3 in the muscle tissue of the IBM group were higher than those of the PM group, the differences were statistically significant (P<0.05). Conclusion TDP-43 and LC3 are more easily expressed in IBM skeletal muscle specimens, and can be used as one of the indicators for differential diagnosis from PM.
