Effect of Wenyang Sanxian Decoction on the contents of hydroxyproline and transforming growth factor-β1 in lung tissue of mice with pulmonary interstitial fibrosis induced by Bleomycin

Abstract:Objective To explore effect of Wenyang Sanxian Decoction on the contents of hydroxyproline (Hyp) and transforming growth factor-β1(TGF-β1)in lung tissue of mice with pulmonary interstitial fibrosis induced by Bleomycin.Methods Bleomycin (0.2-0.3 ml, 5 mg/kg) was injected into C57BL/6 mice through the trachea to establish a pulmonary interstitial fibrosis model. The successful 18 pulmonary fibrosis mice were randomly divided into model group,Wenyang Sanxian Decoction low, medium and high dose groups [5.6625, 11.3250, 22.6500 mg/(kg·d)], positive group[15 mg/(kg·d) Methylprednisolone], with 3 mice in each group. In addition, sham operation and normal group were established, with 3 mice in each group. The contents of Hyp and TGF-β1 in lung tissue were detected by enzyme linked immunosorbent assay (ELISA). Results The contents of Hyp and TGF-β1 in the model group were higher than those in the normal group and sham group, the differences were statistically significant (P<0.05). The contents of Hyp and TGFβ1 in the Wenyang Sanxian Decoction low, medium and high dose groups and positive group were lower than those in the model group, the differences were statistically significant (P <0.05). The contents of Hyp and TGF-β1 in the Wenyang Sanxian Decoction middle, high dose groups and positive group were higher than those in the Wenyang Sanxian Decoction low dose group, the differences were statistically significant (P<0.05). The contents of Hyp and TGF-β1 in the Wenyang Sanxian Decoction high dose group and positive group were lower than those in the Wenyang Sanxian Decoction middle dose group, the differences were statistically significant (P<0.05). Conclusion A certain dose of Wenyang Sanxian Decoction could improve the pulmonary interstitial fibrosis of C57BL/6 mice by reducing the contents of Hyp and TGF-β1.
