Abstract:Objective To investigate the effect of Licorice Chalcone A on the proliferation and apoptosis of human glioma U251 cells.Methods Human glioma U251 cells cultured in vitro were divided into four groups (n=20).The control group was treated with the same amount of 0.9% NaCl,the low-dose Licorice Chalcone A group was treated with 15 μmol/L Licorice Chalcone A,the medium-dose Licorice Chalcone A group was treated with 30 μmol/L Licorice Chalcone A,the high-dose Licorice Chalcone A group was treated with 45 μmol/L Licorice Chalcone A.All were treated for 48 h.The cell proliferation levels of the four groups were detected by methyl thiazolyl tetrazolium colorimetry method.The expression levels of Wnt/β-catenin signaling pathway related proteins β-catenin and Cyclin D1 mRNA in the four groups were detected by real-time fluorescence quantification.The protein expression levels of proliferating nuclear antigen Ki-67 (Ki-67) and B lymphoma-2 (Bcl-2) in four groups were detected by Western blot.Results The cell proliferation levels,β-catenin mRNA,Cyclin D1 mRNA relative expression levels,Ki-67 and Bcl-2 in the lowdose Licorice Chalcone A group,the medium-dose Licorice Chalcone A group and the high-dose Licorice Chalcone A group were lower than those in the control group,the differences were statistically significant (P <0.01).The cell proliferation levels,β-catenin mRNA,Cyclin D1 mRNA relative expression levels,Ki-67 and Bcl-2 in the medium-dose Licorice Chalcone A group and the high-dose Licorice Chalcone A group were lower than those of the low-dose Licorice Chalcone A group,the differences were statistically significant (P<0.01).The cell proliferation levels,β-catenin mRNA,Cyclin D1 mRNA relative expression levels,Ki-67 and Bcl-2 protein relative expression in the high-dose Licorice Chalcone A group were lower than those of the medium-dose Licorice Chalcone A group,and the differences were statistically significant (P<0.01).Conclusion Licorice Chalcone A can inhibit the proliferation and promote apoptosis of human glioma U251 cells.The mechanism may be related to down-regulation of β-catenin mRNA,Cyclin D1 mRNA,Ki-67 and Bcl-2 expression levels.
