Abstract:Objective According to the principle of isolating air and inhibiting bacterial growth, the application effect of vegetable oil as preservative for the preservation of 24 h urine protein quantitative specimens was studied. Methods From November 2020 to September 2021, 30 samples of urine routine and urine visible component analysis in Shenzhen Bao′an Traditional Chinese Medicine Hospital were selected as the research subjects. The semi-quantitative results of urine protein were ±, 1+, 2+, 3+ and 4+ as screening criteria, and 6 samples were selected from each gradient. The samples were divided into experimental group and comparison group with 30 samples in each group. The quantitative results of urine protein in the comparison group were detected by biochemical analyzer and used as the comparison value, In the experimental group, the samples were sealed with vegetable oil as liquid level and isolated from air. The samples were placed at room temperature for 24 h, and the quantitative results of urinary protein were detected by biochemical analyzer again as the test value, the quantitative results of urinary protein were compared between the two groups. Results There was no significant difference in urine protein between experimental group and comparison group(P>0.05). The relative bias was (-1.37±8.25) %, and there was no significant difference between the two groups (P>0.05). According to the scheme in the CLSI guideline EP15-A2 of using patient specimens to verify the accuracy of the two methods, the absolute value of the relative bias of the test group and the comparison group was calculated to be 1.37%, which was less than the 1/2 allowable total error standard used by clinical laboratories, 22%. Conclusion Vegetable oil can be used for 24 h urine protein quantitative sample storage at room temperature, vegetable oil preservatives have the advantages of easy material, economic safety, convenient operation, suitable for popularization.
