Study on optimization of extraction technology and antimicrobial activity of total flavonoids from the aerial part of Bupleurum chinense by orthogonal design

Abstract:Objective To optimize the extraction process of total flavonoids from the aerial part of Bupleurum chinense by orthogonal experiment design and determining its bacteriostatic activity.Methods Based on one factor test,an orthogonal experimental design was employed,with total flavonoids extract ratio as an index,to optimize four extraction factors: solid-liquid ratio,soaking time,extraction time and times of extraction,the antimicrobial activities of four kinds of bacteria and four kinds of fungi were determined by 96-well plate method.Results The optimum technology conditions of extraction total flavonoids from the aerial part of Bupleurum chinense were as follows: ratio of solid to liquid of 1∶10,soaking time of 60 min,extraction time of 72 min,extraction times 3 times.The extraction rate of total flavonoids from the aerial part of Bupleurum chinense was 2.702% by these conditions.The results showed that the total flavonoids in the aerial part of Bupleurum chinense had certain antimicrobial activity against four bacteria and four fungi tested.Conclusion The extraction process is simple,reliable,reasonable and feasible.The total flavonoids in the aerial part of Bupleurum chinense had certain bacteriostatic activity.
