Abstract:Objective To establish a method for detecting Teicoplanin plasma concentrations. Methods Plasma samples were pretreated by acetonitrile precipitation of protein and dichloromethane extraction, and Piperacillin sodium was used as internal standard. Diamonsil C18 chromatographic column (4.6 mm × 150 mm, 5 μm) was used as chromatographic column, acetonitrile and 0.01 mol/L potassium dihydrogen phosphate solution (25∶75) (adjust pH value to 2.8 with phosphoric acid solution) was used as mobile phase, the column temperature was 30℃, the flow rate was 1.2 ml/min, the detection wavelength was 215 nm, the injection volume was 20 μl. Results The peaks of Teicoplanin and internal standard were well shaped with retention times of about 7.3 min and 12.5 min, respectively. Linear regression equation was Y=11.295 386X-0.048 985 (r=0.9987), linear range was 3.125 to 100.000 μg/ml. The intra batch and inter batch accuracy were in the range of 85% to 115%. The relative standard deviation within and between batches was less than 15%. The recoveries of Teicoplanin and internal standard were in the range of 85% to 115%. Conclusion The plasma concentration of Teicoplanin can be determined by high performance liquid chromatography. The method is simple, feasible, specific and reproducible. Methodological indicators are in line with the requirements of Pharmacopoeia of the People′s Republic of China.
