Abstract:Objective To compare the comprehensive bloodletting in different modeling parameters to determine the optimal one with blood deficiency pattern, and to provide data support for the repeatable standardization of the method.Methods A total of 100 ICR mice were randomly divided into 5 groups: normal control group (n=20), model group A(15 min swimming daily+bloodletting 0.3 ml every other day, n=20), model group B (10 min swimming daily+bloodletting 0.3 ml every other day, n=20), model group C (15 min swimming daily+bloodletting 0.5 ml every other day, n=20)and model group D (10 min swimming daily+bloodletting 0.5 ml every other day, n=20). Thymus, spleen and blood were collected for measuring the weight, immune organ index, blood routine indicators (including red blood cells, leukocytes,hemoglobin and platelets), serum interleukin-6 (IL-6) levels and erythrocyte membrane Na+-K+-ATPase activity after nine days model establishment. Results The weight, immune organ index, blood routine indicators (including red blood cells, leukocytes, hemoglobin and platelets), serum IL-6 levels and erythrocyte membrane Na+-K+-ATPase activity in the model group A, B, C, and D were lower than those in the normal control group after nine days model establishment,and the differences were statistically significant (P<0.05). The animal mortality rate of the model group B was lower than that of model group A, C, and D, and the differences were statistically significant (P<0.05). Conclusion Daily swimming for 10 min + bloodletting 0.3 ml every other day is the optimal model parameters for traditional Chinese medicine syndrome of blood deficiency pattern.
