Influence of Polydatin on vascular function in the ApoE -/- mice with atherosclerosis |

Abstract Objective To study the influence of Polydatin on vascular function in the ApoE-/-mice with atherosclerosis.Methods To prepare the mouse model with therosclerosis,16 ApoE-/-mice with high fat diet were selected.The model mice were randomly divided into the model group and the experimental group,and 8 normal mice were chosen as the control group.The experimental group was treated with Polydatin for 16 weeks,while the model group and the control group was not treated.After and before the treatment,the serum was collected,and the triglyceride(TG),low density lipoprotein(LDL-c),tumor necrosis factor α (TNF-α),interleukin 1β (IL-1β),interleukin 6(IL-6),high sensitive C-reactive protein(hs-CRP),total antioxidant capacity(T-AOC),malondialdehyde(MDA),monocyte chemoattractant protein-1(MCP-1),high density lipoprotein(HDL-c),superoxide dismutase(SOD)and interleukin 10(IL-10)were detected by enzyme linked immunosorbent assay(ELISA).The tissue sections of mouse arterial blood vessels were made and the plaque area ratio of arterial blood vessels in each group was compared by oil red O staining.The plaque area ratio of arterial vessels in each group was compared.Results Before the treatment,compared with the control group,the level of TC,TG,LDL-c,TNF-α,IL-1β,IL-6,hs-CRP,T-AOC,MDA,MCP-1 in serum from the model group and the experimental group were significantly increased(P<0.05);the level of HDL-c,SOD and IL-10 of that were significantly reduced(P<0.05).After the treatment,compared with the model group,the level of TC,TG,LDL-c,TNF-α,IL-1β,IL-6,hs-CRP,T-AOC,MDA and MCP-1 in serum from the experimental group were significantly lower,and the HDL-c,SOD and IL-10 increased significantly(P<0.05).And the area of arterial plaque decreased significantly(P<0.05).Conclusion The Polydatin may relieve and prevent certain effect on atherosclerosis in ApoE-/-mice,reduce blood fat,inflammatory cytokines,chemokines,and increase serum immune function.
