Com parative analysis of three testmethods of high density protein cholesterol |

Abstract ObjectiveTo compare and analyze the accuracy and specificity of polyanionic polymer/surfactant(PPD) method and PEG-modified enzyme/sulfateα-cyclodextrin method (PEGME)and precipitationmethod in high-density lipoprotein cholesterol(HDL-C)inspection.MethodsThe known concentration 1.0 to 2.0,2.0 to 4.0 mmol/L of HDL-C, LDL-C were tested on biochemical analyzer,the parameters were analyzed by two point end method,the primary and secondary wavelength were 600 nm,700 nm,respectively,240μl R1 were joint to 3μl sample,288 s absorbance A1volume was readed out,the reaction for 5 min before the 80μl R2 added,600 s absorbance A2after reading it.valueswere calculated its content according to the reference product of HDL-C,LDL-C concentration.ResultsPPD reagent had relatively slow effect on HDL-C,nonspecific response to LDL-C was up to 10%to 20%;PEGME reagent had relatively high effect on HDL-C,the LDL-C did not have a nonspecific response,only 5%.The normal values of HDL-C,LDL-C and 1.1 to 1.7mmol/Lwere respectively Trig,1.7 to 2.9,0.4 to 1.8,and the result of PPD method was higher than that of precipitation method 7%to 15%.The reference set for precipitation results,if the serum samples in normal bilirubin concentration range,so the test results of the PPD method with false increase performance,and if the serum bilirubin concentration of>20 mol/L,then PPD assay results had significantly lower performance,reached the reference value of 60%;the change of bilirubin concentration little interference PEGME determination results.The reference set for precipitation results,PPD determination results were influnced by the triglyceride,and was significantly improved more than 20%,while raising the concentration of triglyceride increased further pseudo PEGMEmethod;less triglyceride effect.The two were negatively correlated with triglyceride concentration.The result of PPD method was significantly higher than that of precipitationmethod (t=4.303,P<0.05).ConclusionThe PEGMEmethod is better than PPDmethod in HDL-C inspection,the clinical laboratory should prudent application of PPDmethod,so isworthy of the clinical′s attention.
