Abstract:ObjectiveTo investigate the effect of astragalus polysaccharide in multiple administrations on humoral,cellular,and nonspecific immune function in mice from day 14 to 17.MethodsMale Kunming mice were selected as the subjects,and randomly divided into 8 groups:blank control group(taking into account that the cellulose itself also belongs to polysaccharide,and blank control group was set in order to exclude its impact),normal control group,model group,1000 mg/kg Ganodermalucidum polysaccharide group,and groups of Astragalus polysaccharide in doses of 125 mg/kg,250 mg/kg,500 mg/kg and 1000 mg/kg.There were 12 animals in each group.Each day,8:00 was for fasting,and 14:00 for intragastric administration based on the weight(0.4 ml/20 g).There was no drug or menstruum in the blank control group.In the normal and model groups,same volume of 0.5%CMC was offered.After 2-hour medication,food was provided with 3 g each mouse.The medication was lasted for 15 days.In day 10 of administration,S180 tumor-bearing cells were inoculated to the right axilla in mice except blank and normal control groups.24 hour after medication,blood was collected from eye socket,and serum was separated.The serum was diluted in 3 times to determine the diameter of IgG precipitation ring.By reference of the standard IgG,the relative content of IgG in the sample was calculated.The experimental results were statistically analyzed.ResultsAstragalus polysaccharide in 125,250,500 mg/kg,and 1000 mg/kg could significantly increase the serum IgG content in the immunosuppressive mice induced by cyclophosphamide and tumorbearing.Astragalus polysaccharide in 500 mg/kg and 1000 mg/kg greatly increased the content of serum IgG in irradiated mice.After skin allergy due to exposure to 2,4-dinitrofluorobenzene,the delayed allergic reaction mediated by T cells could be induced by repeated skin contact about one week.Astragalus polysaccharide in 250 mg/kg and 500 mg/kg could increase delayed allergic reaction in normal mice induced by 2,4-dinitrofluorobenzene.If in 125 mg/kg and 500 mg/kg,astragalus polysaccharidecould increase the delayed allergic reaction in immunosuppressive miceinduced by cyclophosphamide which was from 2,4-dinitrofluorobenzene.Astragalus polysaccharide in 250 mg/kg and 500 mg/kg could obviously restore the phagocytic function in immunosuppressive mice induced by cyclophosphamide,and 500 mg/kg could markedly raise the carbon clearance index K and phagocytic index α in mice.ConclusionIt is indicated that thatastragalus polysaccharide has the function of increasing humoral immunity,promotes the function of T-lymphocyte immune cells,and can significantly improve the nonspecific immune function in mice.
